目的研究根皮苷对体外LO2肝细胞脂肪变性的抑制作用。方法体外培养LO2细胞,实验分正常对照组、模型组、辛伐他汀组和不同剂量的根皮苷组。药物作用24,48,72h后分别检测油红O染色后脂滴吸光度,细胞内甘油三酯(TG)含量和细胞外培养基葡萄糖水平。结果与模型组比较,光镜下各给药组的细胞脂滴均明显减少。在24,48,72h时模型组吸光度均显著高于正常对照组(P<0.01),各给药组吸光度均显著低于模型组(P<0.05,P<0.01);在24,48,72h时模型组TG水平均显著高于正常对照组(P<0.05,P<0.01),辛伐他汀组及根皮苷高、中剂量组TG水平均显著低于模型组(P<0.05,P<0.01);在24h时模型组细胞培养基上清液葡萄糖含量均高于正常对照组(P<0.05),各给药组葡萄糖含量均显著低于模型组(P<0.05,P<0.01)。结论根皮苷对油酸诱导的体外LO2肝细胞脂肪变性和糖代谢异常有显著的抑制作用。
Objective To study the inhibitory effect of phlorizin against fatty degeneration of LO2 hepatocytes in vitro. Methods LO2 cells were cultured in vitro. The experiment was divided into normal control group, model group, simvastatin group and different doses of phlorizin group. After 24, 48 and 72 hours, the lipid droplets absorbance, intracellular triglyceride (TG) content and extracellular medium glucose level were measured respectively after oil red O staining. Results Compared with the model group, the lipid droplets in each group were significantly reduced under the light microscope. At 24h, 48h and 72h, the absorbance of the model group was significantly higher than that of the normal control group (P <0.01), and the absorbance of each group was significantly lower than that of the model group (P <0.05, P <0.01) (P <0.05, P <0.01). TG levels in simvastatin group and phlorizin group were significantly lower than those in model group (P <0.05, P < 0.01). The glucose content in the supernatant of cell culture medium in model group at 24 hours was higher than that in normal control group (P <0.05). The glucose content in each group was significantly lower than that in model group (P <0.05, P <0.01). Conclusion Phloridzin has a significant inhibitory effect on the fatty degeneration and glucose metabolism of LO2 hepatocytes induced by oleic acid in vitro.
