目的探讨中药材鸡冠花中有效部位止血作用机制。方法鸡冠花的正丁醇部位提取物分别以高、中、低剂量给大鼠连续灌胃给药14d后,剥离腹主动脉,测定大鼠活化部分纤维蛋白原(FIB)、凝血酶时间(TT)、凝血活酶时间(APTT)、凝血酶原时间(PT)各项指标,考察大鼠血液凝集参数的变化情况;给小鼠连续灌胃给药7d后,摘除眼球取血,用ELISA法测定纤溶酶原激活物(t-PA)和纤溶酶原激活物抑制物(PAI-Ⅰ)值。结果鸡冠花正丁醇液能够缩短大鼠APTT、PT值,对二者的影响呈剂量正相关性,对大鼠TT、FIB值的影响不显著;能够降低小鼠的t-PA值,提高PAI-Ⅰ值。结论鸡冠花正丁醇提取物具有良好的止血作用,其作用机制可能与缩短APTT、PT值,抑制纤溶酶原激活物活性,提高纤溶酶原激活物抑制物活性有关。
Objective To investigate the mechanism of hemostasis in the effective part of Chinese herbal medicine Celosia. Methods The extract of n-butanol fraction of Clover flower was treated with high, medium and low doses of gavage for 14 days. The abdominal aorta was dissected and the fibrinogen (FIB), thrombin time TT, APTT and PT were measured to investigate the changes of blood coagulation parameters in rats. After mice were given gavage for 7 days, (T-PA) and plasminogen activator inhibitor (PAI-I) were measured by ELISA. Results CIG could shorten the APTT and PT of rats, showing a dose-dependent effect on the two, but had no significant effect on the TT and FIB of rats. It could reduce the t-PA and increase PAI-I value. Conclusion The n-butanol extract of celosia fargesii has a good hemostatic effect. Its mechanism may be related to shortening APTT, PT, inhibiting plasminogen activator activity and increasing plasminogen activator inhibitor activity.
