利用溴和萤光素的卤代反应测定微量溴早有报导。青海盐湖研究所的资料认为在pH5.5醋酸~醋酸钠缓冲溶液中显色后,再加入氢氧化钠增大pH至5.8,可以使红色稳定5分钟。此条件不够理想。我们改为盐酸~醋酸铵缓冲溶液并确定于pH3.8~4.2显色,有良好重现性。显色后加入甲酸钠并加热至60℃~70℃,可破坏残留氧化剂而使红色长期稳定。溴代萤光素能够被正丁醇完全萃取,籍此可以提高分析灵敏度。天然水和盐矿中共存离子均不干扰,氯离子可以加速反应,含量在60~120毫克效果最好。本法适合分析
The use of bromine and luciferin halogenation reaction of trace bromine has been reported. Bromo-Luciferin can be fully extracted with n-butanol, which enhances the sensitivity of the assay.
