海盐县副溶血性弧菌血清分型和毒力基因携带状况分析

目的了解海盐县副溶血性弧菌的优势血清型及毒力基因携带状况。方法收集2015—2016年海盐县腹泻和食物中毒患者(人源株)、食物中毒调查样品(食源株)和海产品(海产株)中分离的96株副溶血性弧菌株,根据GB/T 4789.7—2013《食品微生物检验副溶血性弧菌检验》进行血清分型试验,并采用荧光定量PCR检测耐热直接溶血素基因(tdh)、耐热直接溶血素相关基因(trh)和不耐热直接溶血素基因(tlh)。结果 96株菌株的血清型以O3∶K6和O4∶K8为主,分别占35.42%(34株)和33.33%(32株)。人源株中检出的O3∶K6和O4∶K8型合计占人源株菌株数的87.76%,食源株中检出的占76.67%;海产株中未检出O3∶K6和O4∶K8型。96株菌株tlh基因均阳性,阳性率为100.00%;58株tdh基因阳性,阳性率为60.42%;1株trh基因阳性,阳性率为1.04%。人源株的毒力基因携带率为93.88%,高于食源株的43.33%(P<0.05);海产株中均未检出tdh和trh基因。O4∶K8型菌株毒力基因携带率为87.50%,O3∶K6型菌株毒力基因携带率为76.47%;O3∶K6和O4∶K8型菌株的毒力基因携带率合计为81.82%,高于其他血清型菌株的16.67%(P<0.05)。结论海盐县2015—2016年副溶血性弧菌的优势血清型是O3∶K6和O4∶K8,血清型与毒力基因携带有一定程度关联,O4∶K8血清型毒力基因携带率最高,其次为O3∶K6血清型。 Objective To understand the predominant serotypes and virulence genes of Vibrio parahaemolyticus in Haiyan County. Methods 96 strains of parahaemolyticus isolated from patients with diarrhea and food poisoning (human strain), food poisoning survey sample (food-derived strain) and seafood (marine strain) from 2015 to 2016 were collected according to GB / T 4789.7-2013 “Microbiological examination of food for the detection of Vibrio parahaemolyticus” for serotyping and fluorescence quantitative PCR detection of heat-resistant direct hemolysin gene (tdh), heat-resistant hemolysin-related gene (trh) and impatience Thermal direct hemolysin gene (tlh). Results The serotypes of 96 strains were mainly O3: K6 and O4: K8, accounting for 35.42% (34 strains) and 33.33% (32 strains), respectively. O3: K6 and O4: K8 detected in human strains accounted for 87.76% of the total number of human strains and 76.67% of those detected in the source strains. O3: K6 and O4: K8 were not detected in the marine strains type. 96 strains of tlh gene were positive, the positive rate was 100.00%; 58 tdh gene positive, the positive rate was 60.42%; a trh gene was positive, the positive rate was 1.04%. The virulence rate of human strains was 93.88%, which was higher than 43.33% (P <0.05) of the original strains. No tdh and trh genes were detected in the marine strains. The virulence rate of O4: K8 strain was 87.50%, that of O3: K6 strain was 76.47%, that of O3: K6 strain and O4: K8 strain was 81.82% Other serotype strains of 16.67% (P <0.05). Conclusion The predominant serotypes of Vibrio parahaemolyticus from 2015 to 2016 in Haiyan County are O3: K6 and O4: K8, and the serotypes are associated with virulence genes to a certain extent. O4: K8 serotype virulence genes carry the highest rates, followed by O3: K6 serotype.
