生长因子通过膜受体可将胞外信号转导至胞内 ,现已发现一些小分子肽能够作为受体的配体模拟天然配体的生物学作用。以可溶性受体的纯化蛋白或转染了膜受体的完整细胞为靶标 ,人们从噬菌体肽库中筛选出多种模拟肽 ,对这些肽的序列和生物学功能进行分析 ,发现它们能够竞争受体与天然配体相结合 ,可作为生长因子受体的拮抗剂或激动剂。噬菌体展示技术的应用不仅为生物大分子间的结构和功能关系提供信息 ,而且为新药的研究提供了新思路。本文对从噬菌体肽库中筛选生长因子受体肽类拮抗剂或激动剂的研究进展作一综述。
Growth factors transduce extracellular signals into the cell through membrane receptors. It has been found that some small peptides can mimic the biological role of natural ligands as ligands for receptors. Targeting purified proteins from soluble receptors or whole cells transfected with membrane receptors, many peptides were screened from the phage peptide library. The sequence and biological functions of these peptides were analyzed and found to be competitive The body binds to natural ligands and acts as an antagonist or agonist of the growth factor receptor. The application of phage display technology not only provides information for the relationship between structure and function of biological macromolecules, but also provides new ideas for the research of new drugs. This review summarizes the progress in the screening of phage peptide libraries for antagonists or agonists of growth factor receptor peptides.
