目的 :探讨复制缺陷型腺病毒介导的 p16基因转染 ,对人膀胱癌细胞中增殖细胞核抗原 (PCNA)和P5 3蛋白表达的影响。 方法 :将 p16基因重组腺病毒 (Ad p16 )感染P16蛋白表达缺失的人膀胱癌T2 4细胞株 ,流式细胞术评估Ad p16对T2 4细胞中PCNA和P5 3蛋白表达的影响。 结果 :PCNA主要在S期和G2 ~M期高表达 ,Ad p16转染T2 4细胞后 ,PCNA阳性细胞百分率降低 ,PCNA表达的荧光强度减弱 ;Ad p16转染未能使T2 4细胞表达P5 3蛋白。 结论 :PCNA在Ad p16转染后其含量减少 ,PCNA的表达与细胞的增殖能力密切相关 ,PCNA可作为评估肿瘤细胞恶性增殖能力的指标 ;Ad p16转染不能使T2 4细胞表达P5 3蛋白。
Objective: To investigate the effect of replication-deficient adenovirus-mediated transfection of p16 gene on the expression of proliferating cell nuclear antigen (PCNA) and P53 protein in human bladder cancer cells. Methods: Human bladder cancer T2 4 cell line with P16 protein expression was infected by recombinant adenovirus p16 gene. The effect of Ad p16 on the expression of PCNA and P53 protein in T2 4 cells was evaluated by flow cytometry. Results: PCNA was mainly expressed in S phase and G2 ~ M phase. The percentage of PCNA positive cells decreased and the fluorescence intensity of PCNA decreased after Ad p16 transfected into T2 4 cells. Ad p16 transfection failed to express P53 protein. CONCLUSION: The expression of PCNA is decreased after Ad p16 transfection and the expression of PCNA is closely related to the proliferation of cells. PCNA can be used as a marker to evaluate malignant proliferation of tumor cells. Ad p16 transfection can not express P53 protein in T2 4 cells.
