为了获得对家蚕核型多角体病毒(BmNPV)感染抵抗力较强的家蚕品系,以现行推广杂交组合粤蚕6号和两广二号的8个亲本品种为材料,给3龄起蚕添食浓度为4× 107 mL-1的BmNPV多角体悬液,连续12代接种病毒筛选后获得7个对BmNPV的抵抗能力有显著提高的新品系,其中新品系研7-N感染BmNPV的发病率由原品种的96.30%降低至51.46%,表现出极显著差异(P<0.01).采用荧光定量PCR方法,检测新品系及其杂交组合幼虫中肠组织中与BmNPV抗性相关的脂肪酶1(Bmlipase-1)基因的表达水平,结果显示不同新品系间Bmlipase-1基因的表达水平差异较大,与其对BmNPV的抵抗能力一致,其中春5-N、研7-N和湘晖-N等新品系及其杂交组合的Bmlipase-1表达水平均较原品种及其杂交组合极显著上升(P<0.01).获得的家蚕新品系可作为抗BmNPV育种的优良素材.“,”In order to obtain silkworm (Bombyx mori) strains resistant to B.mori nucleopolyhedrovirus (BmNPV),we took 8 parental silkworm varieties of currently popularized hybrid combinations Yuecan No.6 and Liangguang No.2 as materials,fed the 3rd instar newly exuviated larvae with mulberry leaves treated by BmNPV polyhedron suspension at concentration of 4×107 mL-1 for 12 generations successively.7 new silkworm strains with significantly increased resistance to BmNPV were obtained,of which BmNPV infection rate of Yan 7-N was reduced from 96.30% (the original strain) to 51.46% (the newly bred strain),which showed extremely significant difference (P<0.01).The expression level of Bmlipase-1 in midgut of the newly bred strains and their hybrid combinations was detected by fluorescence quantitative PCR.The results showed that the expression level of Bmlipase-1 varied greatly among different varieties,being consistent with their resistance to BmNPV.Expression of Bmlipase-1 in newly bred strains Chun 5-N,Yan 7-N,Xianghui-N and their hybrid combinations was extremely significant higher than that of the original strains and their hybrid combinations (P<0.01).The newly bred silkworm strains can be used as excellent materials for anti-BmNPV breeding.
