目的对42个常见的肿瘤细胞株进行筛选,明确细胞株中是否存在CD133+可能的肿瘤干细胞。方法取对数生长期的肿瘤细胞株,CD133抗体标记后用流式细胞仪检测,观察CD133+细胞的比例。结果 42个肿瘤细胞株中,肝癌细胞株SK-HEP-1、HepG2、Hep3B、L2-2-15、PLC/PRF5,胰腺癌细胞株PANC-1、BXPC-1,结肠癌细胞株SW620、HT29,鼻咽癌细胞株CNE-2,共10个细胞株CD133+细胞数与对照组相比差异有统计学意义(P<0.01)。结论 42个肿瘤细胞株中检出10个细胞株存在CD133+可能的肿瘤干细胞亚群。
Objective To screen 42 common tumor cell lines and determine whether there are CD133 + potential cancer stem cells in the cell lines. Methods Tumor cell lines in logarithmic growth phase were stained with CD133 antibody and detected by flow cytometry to observe the proportion of CD133 + cells. Results Among the 42 tumor cell lines, the hepatocellular carcinoma cell lines SK-HEP-1, HepG2, Hep3B, L2-2-15, PLC / PRF5, pancreatic cancer cell lines PANC-1, BXPC-1, colon cancer cell lines SW620, HT29 (P <0.01). The number of CD133 + cells in 10 nasopharyngeal carcinoma cell lines CNE-2 cells was significantly higher than that of the control group (P <0.01). Conclusions CD133 + may be a subset of tumor stem cells in 10 out of 42 tumor cell lines.
