以pSM889为探针,对分布于中国大陆安徽、湖北、广西、云南、四川5地日本血吸虫自然隔离群的雄虫DNA进行了杂交,分析其长度多态性差异。经限制性内切酶EcoRI和BamHI酶切的杂交图谱分析,结果表明其强杂交带虽相同,但在EcoRI酶切的杂交图谱中,分子量分别在4.4-9.6kb和2.3-4.4kb的上述5地隔离群的弱杂交带上却呈现明显的差别。这表明5地自然隔离群日本血吸虫的DNA结构发生了某些变异,此结果进一步为中国大陆境内日本血吸虫存在品系差别提供了分子生物学的依据。
Using pSM889 as a probe, the male DNAs of Schistosoma japonicum natural isolates distributed in Anhui, Hubei, Guangxi, Yunnan and Sichuan in mainland China were crossed and their differences in length polymorphism were analyzed. The result of restriction endonuclease digestion with EcoRI and BamHI showed that the strong hybridization bands were the same in the EcoRI digested hybridization map with the molecular weights of 4.4-9.6kb and 2.3-4.4kb respectively Weak hybridization to the isolation group has shown a significant difference. This indicates that there are some variations in the DNA structure of Schistosoma japonicum in five natural isolates. This result further provides the molecular biology basis for the existence of the strain of Schistosoma japonicum in mainland China.
