目的:分析趋化因子CXCL12及其受体CXCR4在人类精子的表达。方法:收集有生育史且精液常规分析结果正常男性精液标本10例,经过密度梯度离心纯化处理后,采用RT-PCR和免疫荧光化学染色技术在基因和蛋白水平分析CXCL12/CXCR4在人精子的表达。结果:虽然正常人精子同时表达CXCL12(0.641±0.180)与CXCR4(0.464±0.100)mRNA,但免疫荧光染色分析发现,在蛋白水平人精子主要表达CXCR4而不表达CXCL12,且CXCR4染色主要位于精子顶体后区。结论:CXCL12/CXCR4可能也是调节精子功能的重要分子。
Objective: To analyze the expression of chemokine CXCL12 and its receptor CXCR4 in human sperm. Methods: Ten samples of male semen with reproductive history and routine semen analysis were collected. After density gradient centrifugation, the expression of CXCL12 / CXCR4 in human sperm was analyzed by RT-PCR and immunofluorescence staining at the gene and protein levels . Results: Although normal human sperm expressed both CXCL12 (0.641 ± 0.180) and CXCR4 (0.464 ± 0.100) mRNA, immunofluorescence staining showed that CXCR4 was mainly expressed in human sperm at the protein level but not CXCL12, and CXCR4 staining was mainly located at the spermoptima Body area. Conclusion: CXCL12 / CXCR4 may also be important molecules regulating sperm function.
