兔颈动脉颈静脉旁路手术后 ,将阳离子脂质体 DOSPER介导的 p5 3腺相关病毒增强型质粒表达载体 ,经腔内加压灌注转染移植静脉. RT PCR和免疫组化检测证明 ,外源性 p5 3基因在移植血管中表达出相应的 m RNA及 p5 3蛋白.结果提示 :腔内加压灌注是移植静脉基因转移的有效方法之一“,”The purpose of this study is to develop a new way to transfer extrinsic gene to vein graft.The models of carotidjugular bypass were prepared in rabbits.The human wildtype p53 gene adenoassociated virus plasmid vector was transfected into vein graft mediated by the cationic liposome DOSPER by using intraluminal pressure perfusion method.The gene expression was determined by polymerase chain reaction technique and immunohistochemistry.It was verified that the extrinsic p53 gene was transduced into vein graft successfully and expressed mRNA and p53 protein.The results suggested that intraluminal pressure perfusion method is one of the efficient means for vein autograft gene transfer system.
