目的 :运用基因表达谱芯片技术对肿瘤转移相关基因的表达谱进行研究。方法 :对临床切除的肝癌和胰腺癌组织及相应的对照正常组织进行总 RNA抽提 ,纯化后的 m RNA进行逆转录制备杂交探针 ,应用 Bio Door40 96和 Bio Door12 80 0的全长基因 DNA芯片筛选与肝癌和胰腺癌转移相关的基因。杂交后的信号用 Scan Array30 0 0扫描 ,结果用 Ima Gene3.0软件分析。结果 :对肝癌和胰腺癌基因表达谱进行分析 ,发现两种肿瘤中存在相同的差异表达基因 ,其中有 15条基因与肝癌、胰腺癌转移密切相关。 结论 :通过对肝癌、胰腺癌的肿瘤表达谱芯片研究 ,尤其是与转移相关基因的分析 ,对了解肿瘤的发病机制和转移发生情况及其肿瘤的预后有重要意义。
Objective: To study the gene expression profile of tumor metastasis genes using gene expression microarray technology. METHODS: Total RNA was extracted from clinically resected HCC and pancreatic cancer tissues and the corresponding normal control tissues. The purified m RNA was used for reverse transcription to prepare hybridization probes. The full-length DNA of Bio Door 40 96 and Bio Door 1880 was used. The microarray screens for genes involved in the metastasis of liver cancer and pancreatic cancer. The hybridized signal was scanned with a Scan Array 3000, and the results were analyzed using Ima Gene 3.0 software. Results : The gene expression profiles of HCC and pancreatic cancer were analyzed. It was found that there were the same differentially expressed genes in the two tumors. Among them, 15 genes were closely related to the metastasis of liver cancer and pancreatic cancer. Conclusion : The study of tumor expression profile of liver cancer and pancreatic cancer, especially the analysis of metastasis-related genes, is of great significance for understanding the pathogenesis, metastasis and prognosis of tumors.
