本文从卵巢上皮癌腹水免疫复合物中,解离出患者的自身抗原(OC_(41))免疫小鼠,取其脾细胞用于融合,经ELISA、PAP及流式细胞计等方法筛选和鉴定,获得2株杂交瘤细胞系,分别命名为COC_(41)D和COC_(41)E,2种细胞诱生小鼠腹水,ELISA测定抗体滴度达2×10~5和1×10~5。免疫组化分析表明,COC_(41)D和COC_(41)E与卵巢上皮癌具有较高的特异性,阳性率分特为70.2％和52.5％。本研究表明,应用腹水免疫复合物解离抗原有可能获得特异性较高的单克隆抗体。
In this study, we immunized mice with self-antigen of patients (OC_ (41)) from the ascites immune complex of ovarian epithelial carcinoma and selected the spleen cells for fusion. The results were screened and identified by ELISA, PAP and flow cytometry , Two hybridoma cell lines were obtained and named as COC_ (41) D and COC_ (41) E respectively. The ascites of mice were induced by two kinds of cells. The titer of antibody was 2 × 10 ~ 5 and 1 × 10 ~ 5 . Immunohistochemical analysis showed that COC_ (41) D and COC_ (41) E were highly specific to ovarian epithelial carcinoma with positive rates of 70.2% and 52.5%, respectively. This study shows that it is possible to obtain monoclonal antibodies with high specificity by dissociating antigens with ascitic immune complexes.
