目的应用闭合式颅骨开窗法,应用激光多普勒微循环血流仪,观察葛根素、川芎嗪、丹参注射液对脑微循环的影响。方法 Wistar大鼠 50只,腹腔注射 1%戊巴比妥钠 40mg/kg麻醉,将动物固定于大鼠立体定位仪上,于右侧领顶部用牙科钻钻开一直径为 6×8mm大小的颅窗,彻底止血后,小心剪开硬脑膜,暴露软脑膜,玻璃及牙科水泥覆盖密闭,将激光多普勒微循环血流计的激光探头固定于颅窗上。之后动物取侧卧位固定,将右侧颈总动脉提起,结扎近心端,小心剪开颈总动脉,将一直径约0.3mm的尼龙线,穿入动脉,并同时观察激光多普勒微循环血流分析仪所显示的脑微循环血流量,穿到大脑中动脉时可出现微循环灌流量的突然下降,出现微循环血流量下降后,再向内穿入约1mm,然后将切口的远心端及动脉内的丝线牢固结扎,剪去多余丝线。实验结束后,查看尼龙线是否阻塞大脑中动脉的起始部,未阻塞动脉者实验数据作废。大鼠随机分为葛根素组、丹参注射液组、川芎嗪组、生理盐水组、空白对照组。结果阻断大脑中动脉后,供血区(额顶叶)脑微循环血流量迅速下降,并维持一较低水平,至阻断后50min脑微循环血流量才有所增加,但增加幅度较低.说明此MCAO模型是成功的。各用药组用药后脑微
Objective To investigate the effect of puerarin, tetramethylpyrazine, and salvia miltiorrhiza injection on cerebral microcirculation by using closed-door skull window method and laser Doppler microcirculatory blood flow meter. Methods 50 Wistar rats were anesthetized by intraperitoneal injection of 1% pentobarbital sodium 40mg/kg. The animals were fixed on the rat stereotaxic apparatus. A diameter of 6×8mm was drilled with a dental drill on the top of the right side collar. The cranial window, after the hemostasis was completely stopped, the dura was carefully cut, the pia mater was exposed, and the glass and dental cement cover were closed. The laser probe of the laser Doppler microcirculation blood flow meter was fixed on the cranial window. After the animal was fixed in the lateral position, the right common carotid artery was lifted, the proximal end was ligated, and the common carotid artery was carefully cut. A nylon thread with a diameter of about 0.3 mm was inserted into the artery and the laser Doppler was observed at the same time. The microcirculatory blood flow in the brain revealed by the microcirculatory blood flow analyzer can cause a sudden drop in the microcirculatory perfusion flow when passing through the middle cerebral artery.
