3B9单抗是一株抗HBV PreS2抗原的单抗,为从分子水平分析,了解389单抗进而为制备新一代基因工程抗体奠定基础。本文用分子生物学技术,提取389杂交瘤细胞总RNA,经反转录,PCR分离获得了389单抗的轻,重链可变区基因。并利用PCR及双脱氧核苷酸链末端终止法对389单抗的可变区基因的核苷酸序列进行了分析。结果表明,389单抗轻链隶属小鼠Ig Kappa轻链第
3B9 monoclonal antibody is a monoclonal antibody against HBV PreS2 antigen, which is to understand the 389 mAb from the molecular level to lay the foundation for the preparation of a new generation of genetically engineered antibodies. In this paper, total RNA was extracted from 389 hybridoma cells using molecular biology techniques. The light and heavy chain variable region genes of 389 mAb were obtained by reverse transcription and PCR. The nucleotide sequence of the variable region gene of 389 mAb was analyzed by PCR and dideoxynucleotide chain termination. The results showed that the 389 mAb light chain belongs to the mouse Ig Kappa light chain
