本文采用22厘米长管和一定比例的Ampholyte等电聚焦—交叉电泳和等电聚焦—交叉电泳放射自显影术分析了人体胎儿血清和肝癌患者血清AFP的变种。胎儿AFP可以有十个变种,pI值分别为AFP_1 4.70,AFP_2 4.85,AFP_3 5.12,AFP_4 5.28,AFP_5 5.48,AFP_6 5.62,AFP_7 5.76,AFP_8 5.81,AFP_9 5.95和AFP_10 4.56。原发性肝癌血清的AFP仅四个变种,分别相当于胎儿的AFP_1,AFP_2,AFP_5和AFP_6。此外还观察了AFP_(1—5)各变种对神经氨酸酶的敏感性、热失活的动力学行为等,彼此性质并不一致。根据上述结果,本文认为AFP各变种不是假象,并对可能的应用前景进行了讨论。
In this paper, 22 cm long tube and a certain proportion of Ampholyte isoelectric focusing - cross electrophoresis and isoelectric focusing - cross-electrophoresis autoradiography were used to analyze the serum AFP variants in human fetal serum and liver cancer patients. There are ten variants of fetal AFP with pI values of AFP_1 4.70, AFP_2 4.85, AFP_3 5.12, AFP_4 5.28, AFP_5 5.48, AFP_6 5.62, AFP_7 5.76, AFP_8 5.81, AFP_9 5.95 and AFP_10 4.56. Only four variants of AFP in serum of primary liver cancer are equivalent to fetal AFP_1, AFP_2, AFP_5 and AFP_6. In addition, the sensitivity of AFP_(1-5) variants to neuraminidase and the kinetic behavior of heat inactivation were also observed. Based on the above results, this paper believes that the AFP variants are not illusions and discuss possible application prospects.
