一、目的与材料方法 红菜薹属白菜类(B.campestris),其原生质体培养成功可供外源基因的直接转化和细胞质杂种合成雄性不育系之用。白菜类是十字花科中最难由原生质体再生出植株的种类之一(Hachey等,1991)。目前,大白菜的再生成株已有报道(李世君等,1992;Glimelius,1984),但再生频率低(3.3%,1%)。本试验试图建立起红菜薹原生质体有效的再生程序。 1.无菌苗的获得 红菜薹品种为十月红一号,种子在0.1%SDS的0.1%HgCl_2溶液中浸5分钟后,用15%的“Domestos”灭菌15分钟,再用无菌水洗5次,播种在MS基本培养基上发芽,25℃,光照每天16小时,光强1800lx。

At present, regenerated plants of Chinese cabbage have been reported (Li Shijun et al., 1992; Glimelius, 1984), but the regeneration frequency is low (3.3% and 1%). This experiment attempted to establish a protoplast of red flowering sedge effective regeneration program. 1. Obtained aseptic seedlings Red flowering sesame varieties Red October, 0.1% SDS in 0.1% HgCl2 solution immersed in 5 minutes, with 15% of the “Domestos” sterilized for 15 minutes, then sterilized Washed 5 times, sown on MS basal medium germination, 25 ℃, light 16 hours a day, light intensity 1800lx.
