采用石炭酸品红染色法刮片检查56名正常女性的睑结膜、颊粘膜和唇粘膜。按常规计算其 X 染色质。结果是睑结膜的发现率最高。40名正常女性睑结膜刮片,在每一个正常结膜细胞核内都有一着色特别深的斑点,可位于细胞核内各处或贴核膜。而在60名正常男性的刮片上却没有着色特别深的斑点。因此推论在正常女性体细胞间期,没有活性的 X 染色体是贴核膜的,但由于核象球形,在制片上 X 染色质可见于核内各处。这一证据接受了 Lyon 假说。
