目的:观察人脐血干细胞(hUCBSC)经静脉移植对心肌梗死(MI)大鼠血管新生和血管内皮生长因子(VEGF)、促血管生成素(Ang-1)表达的影响。方法:雄性SD大鼠40只,随机分为3组:①假手术组(10只);②对照组(15只)制备MI模型成功后,经尾静脉注射0.9%氯化钠溶液0.5 ml;③移植组(15只)尾静脉注射hUCB-SC 0.5 ml(含干细胞2×107/ml)。术后4周行超声心动图和血流动力学检查,并取心脏组织行苏木精-伊红染色、抗Ⅷ因子免疫组化染色,观察心肌病理改变、心肌毛细血管密度(MCD)的变化。同时用免疫组化和RT-PCT的方法检测VEGF、Ang-1及其mRNA的表达。结果:与对照组相比,术后4周时移植组心功能明显改善(P<0.01),左室舒张末压显著降低[(20.13±5.63)mmHg∶(10.56±4.69)mmHg,P<0.01],而左室压力最大变化率明显增加[(4.25±2.01)mmHg/ms∶(6.53±2.38)mmHg/ms,P<0.05];和[(3.83±2.69)mmHg/ms∶(6.25±2.92)mmHg/ms,P<0.05];移植组MCD明显高于对照组[(4.2±0.5)个/HP:(2.1±0.3)个/HP,P<0.01];移植组VEGF和Ang-1及其mRNA表达明显高于对照组和假手术组。结论:hUCBSC经静脉移植通过促进VEGF和Ang-1表达而促进MI大鼠血管新生,从而改善MI大鼠心功能。
Objective: To observe the effects of human umbilical cord blood stem cells (hUCBSC) transplantation on angiogenesis and the expression of vascular endothelial growth factor (VEGF) and angiopoietin (Ang-1) in rats with myocardial infarction (MI) Methods: Forty male Sprague-Dawley rats were randomly divided into three groups: ① sham operation group (n = 10); ② control group (n = 15). After MI model was successfully established, 0.9% sodium chloride solution ③ transplantation group (15) tail vein injection of hUCB-SC 0.5 ml (including stem cells 2 × 107 / ml). Echocardiography and hemodynamics were performed 4 weeks after operation. The heart tissues were harvested for hematoxylin-eosin staining and anti-Ⅷ factor immunohistochemical staining. The changes of myocardial pathological changes, myocardial capillary density (MCD) . Meanwhile, the expressions of VEGF, Ang-1 and its mRNA were detected by immunohistochemistry and RT-PCR. Results: Compared with the control group, the cardiac function of the transplantation group was significantly improved at 4 weeks after operation (P <0.01), and the left ventricular end-diastolic pressure was significantly lower than that of the control group [(20.13 ± 5.63) mmHg: (10.56 ± 4.69) mmHg ], While the maximum rate of change of left ventricular pressure was significantly increased [(4.25 ± 2.01) mmHg / ms: (6.53 ± 2.38) mmHg / ms, P <0.05] ) mmHg / ms, P <0.05]. The MCD in the transplantation group was significantly higher than that in the control group [(4.2 ± 0.5) /HP:(2.1 ± 0.3) /HP, P <0.01] The mRNA expression was significantly higher than the control group and sham operation group. CONCLUSION: HUCBSC can improve angiogenesis in MI rats by promoting the expression of VEGF and Ang-1 after intravenous transplanting, thus improving heart function in MI rats.
