探讨IL 10抑制肾病患儿外周血单个核细胞 (PBMC )培养上清液对大鼠肾小球上皮细胞 (GEC )粘附的影响。应用同位素标记的方法检测GEC粘附率。结果显示 ,未治疗组初发肾病患儿PBMC上清液显著降低GEC粘附率 ,与激素治疗组及正常对照组比较 ,差异有非常显著性意义 (P值均 <0 0 1) ;人重组IL 10明显减弱肾病极期患儿PBMC上清液对GEC粘附的抑制性影响 ,GEC粘附率显著升高 ,并呈剂量依赖效应。表明肾病患儿PBMC产物抑制GEC对胶原基质的粘附 ,而IL 10能负向调节此抑制过程
To investigate the effect of supernatant of peripheral blood mononuclear cells (PBMCs) from children with IL-10 in nephropathy on glomerular epithelial cells (GEC) adhesion. Application of isotope labeling method to detect GEC adhesion rate. The results showed that PBMC supernatant of untreated children with nephropathy significantly reduced the adhesion rate of GEC, compared with the hormone therapy group and the normal control group, the difference was significant (P <0.01); human recombinant IL-10 significantly attenuated the inhibitory effect of PBMC supernatant on the adhesion of GEC in children with advanced stage renal disease. The adhesion rate of GEC was significantly increased in a dose-dependent manner. This suggests that PBMC production in children with nephropathy inhibits GEC adhesion to the collagen matrix, whereas IL-10 negatively regulates this inhibition
