目的 :比较内毒素 (脂多糖 ,LPS)对老龄大鼠和D_半乳糖 (D_gal)致拟衰老大鼠的肝毒性。方法 :给大鼠腹腔内注射 (ip)单一剂量LPS(E .coli,O1 1 1 B4 株 ) ,复制肝损伤。用自动生化仪测定血浆丙氨酸氨基转移酶 (ALT)、天冬氨酸氨基转移酶 (AST)、乳酸脱氢酶 (LDH)、碱性磷酸酶 (ALP)和白蛋白 (Alb) ;光镜观察肝脏病理组织学损伤。结果 :LPS 10mg kgip后 2 4h ,老龄大鼠血浆ALT ,AST ,LDH和ALP活性的升高幅度 (与对照组比 ,分别平均升高2 4 .7,9.0 ,13.3和 4 .2倍 ) ,显著大于D_gal致拟衰老大鼠 (与对照组比 ,分别平均升高 5 .6 ,1.6 ,4 .0和 0 .2倍 ) ;前者肝病理形态学损伤也比后者严重。结论 :LPS对两种衰老大鼠的肝毒性存在较大差别 ,其对老龄大鼠的肝毒性明显大于D_gal致拟衰老大鼠。
OBJECTIVE: To compare the hepatotoxicity of endotoxin (lipopolysaccharide, LPS) in aged rats and D_gal induced aging mice. Methods: Rats were injected intraperitoneally with a single dose of LPS (E.coli, O1 1 1 B4 strain) to replicate liver injury. Plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and albumin (Alb) were measured by automatic biochemical analyzer. Microscopic observation of liver histopathological damage. RESULTS: The levels of ALT, AST, LDH and ALP in the plasma of aged rats increased by 24.7, 9.0, 13.3 and 4.2 times higher than that of the control group respectively Was significantly greater than D_gal induced senile rats (compared with the control group, respectively, an average increase of 5.6, 1.6, 4.0 and 0.2 times); the former liver pathological morphological damage than the latter. CONCLUSION: LPS has significant difference in hepatotoxicity between the two aging rats, and its hepatotoxicity in aged rats is obviously greater than that in D_gal induced aged rats.
