目的探讨奥曲肽对人脐静脉内皮细胞(HUVECs)体外构建新生血管的抑制作用。方法应用噻唑蓝(MTT)实验、流式细胞仪、侵袭和迁移实验、Matrigel实验,研究不同浓度奥曲肽(1×10-10~1×10-5mol/L)对血管内皮生长因子(VEGF)诱导的HUVECs增殖、侵袭、迁移和分化成血管能力的影响。应用逆转录聚合酶链反应(RTPCR)检测HUVECs生长抑素受体(SSTR)的表达。结果奥曲肽浓度在1×10-8~1×10-5mol/L时抑制VEGF诱导的HUVECs增殖;流式细胞仪检测表明HUVECs被扣留在细胞周期G0/G1期;浓度为1×10-7~1×10-5mol/L时,奥曲肽抑制VEGF诱导的HUVECs侵袭作用,对HUVECs迁移无影响;浓度为1×10-8mol/L时,奥曲肽抑制HUVECs体外分化成管样结构,浓度为1×10-6mol/L时,完全阻断HUVECs体外分化成管样结构。RTPCR显示HUVECs表达高水平的SSTR3。结论奥曲肽对HUVECs体外构建新生血管具有抑制作用。
Objective To investigate the inhibitory effect of octreotide on angiogenesis of human umbilical vein endothelial cells (HUVECs) in vitro. Methods The effects of different concentrations of octreotide (1 × 10-10 ~ 1 × 10-5 mol / L) on the induction of vascular endothelial growth factor (VEGF) were investigated by MTT assay, flow cytometry, invasion and migration assay and Matrigel assay Effects of HUVECs proliferation, invasion, migration and differentiation into vascular capacity. The expression of somatostatin receptor (SSTR) in HUVECs was detected by reverse transcriptase polymerase chain reaction (RTPCR). Results The octreotide concentration of 1 × 10-8 ~ 1 × 10-5mol / L inhibited the proliferation of HUVECs induced by VEGF. Flow cytometry showed that HUVECs were arrested at the G0 / G1 phase of the cell cycle. The concentration of octreotide was 1 × 10-7 ~ 1 × 10-5mol / L, octreotide inhibited the invasion of HUVECs induced by VEGF and had no effect on the migration of HUVECs. When the concentration was 1 × 10-8mol / L, octreotide inhibited the differentiation of HUVECs into tube-like structure at the concentration of 1 × 10 -6mol / L, completely blocked HUVECs differentiated into tube-like structure in vitro. RTPCR showed that HUVECs express high levels of SSTR3. Conclusion Octreotide has an inhibitory effect on the construction of neovascular HUVECs in vitro.
