目的 :观察氧应激对豚鼠心肌细胞线粒体的影响 ,探讨抗心肌缺血中药复方———益心康胶囊(H30 3)含药血清对氧应激损伤的保护作用机制。方法 :利用激光共聚焦显微镜 ,观察羟自由基损伤体系(Fe2 + /H2 O2 )与豚鼠心肌细胞共同孵育 30~ 6 0min后对心肌细胞线粒体膜电位的影响。结果 :豚鼠心肌细胞氧应激后 ,可致线粒体细胞色素氧化酶活性降低 ,以及荧光染料Rh12 3所显示的线粒体膜电位明显降低。H30 3含药血清可明显减轻上述损伤 ,表现为明显增强豚鼠心肌细胞线粒体氧化酶活性 ,使心肌细胞存活率增加 ;并具有保护心肌细胞膜电位的作用。结论 :氧应激可造成心肌线粒体膜电位降低。益心康胶囊含药血清可保护线粒体膜电位 ,从而发挥其膜稳定作用。
Objective : To observe the effect of oxygen stress on mitochondria of guinea pig myocardial cells and to explore the mechanism of protective effect of serum containing Yixinkang capsule (H30 3) on oxidative stress injury. METHODS: The effects of hydroxyl radical injury system (Fe2 + /H2 O2) and guinea pig cardiomyocytes on the mitochondrial membrane potential after 30-60 min incubation were observed by confocal laser scanning microscopy. Results : The mitochondrial cytochrome oxidase activity was decreased after oxidative stress in guinea pig cardiomyocytes, and the mitochondrial membrane potential of the fluorescent dye Rh12 3 was significantly reduced. H30 3 drug-containing serum can significantly reduce the above-mentioned damage, manifested as markedly enhanced mitochondrial oxidase activity of myocardial cells in guinea pigs, increased the survival rate of myocardial cells, and has the effect of protecting myocardial cell membrane potential. Conclusion: Oxygen stress can cause a decrease in myocardial mitochondrial membrane potential. Yixinkang capsule-containing serum can protect the mitochondrial membrane potential, thus exerting its membrane stability.
