目的:研究妇科再造胶囊对多囊卵巢综合征(PCOS)患者miRNA-383和miRNA-320表达的影响。方法:选择2014年5月至2015年10月在本院诊断为PCOS患者80例,随机分为对照组和观察组(各40例),对照组采用炔雌醇环丙孕酮片治疗,观察组采用妇科再造胶囊治疗,治疗前和治疗6个月后采集患者血清测定睾酮(TSTO)、黄体生成素(LH)、卵泡刺激素(FSH)含量,采集卵泡颗粒细胞测定miR-383和miR-320的表达量。结果:两组治疗6个月后血清中TSTO、LH含量显著低于治疗前,FSH含量显著高于治疗前,组间比较差异无统计学意义。对照组治疗6个月后颗粒细胞中miR-383和miR-320的表达量无改变;观察组显著降低,差异有统计学意义(P<0.05)。结论:妇科再造胶囊治疗PCOS的可能分子靶点是卵泡颗粒细胞miR-383和miR-320。
Objective: To investigate the effects of gynecological reconstructive capsules on the expression of miRNA-383 and miRNA-320 in patients with polycystic ovary syndrome (PCOS). Methods: 80 patients diagnosed as PCOS in our hospital from May 2014 to October 2015 were randomly divided into control group and observation group (40 cases each). The control group was treated with ethinylestradiol and cyproterone tablets The gynecological reconstructive capsules were used to treat the patients. Serum levels of testosterone (TSTO), luteinizing hormone (LH) and follicle stimulating hormone (FSH) were measured before and 6 months after treatment. The granulosa cells were collected for the determination of miR-383 and miR- 320 expression level. Results: The levels of TSTO and LH in serum of the two groups after 6 months of treatment were significantly lower than those before treatment, and the levels of FSH in the two groups were significantly higher than those before treatment. There was no significant difference between the two groups. In the control group, the expression of miR-383 and miR-320 in the granulosa cells did not change after 6 months of treatment; the observation group was significantly reduced, the difference was statistically significant (P <0.05). Conclusion: Gynecological reconstitution capsules for the possible molecular targets of PCOS are granulosa cells of granulosa cells miR-383 and miR-320.
