目的考察低剂量卡铂诱导人肺癌细胞A549过早性衰老的作用。方法 A549细胞经不同浓度卡铂处理,采用MTT法和克隆形成率试验检测卡铂对细胞增生的抑制作用;β-半乳糖苷酶染色试验检测卡铂诱导细胞衰老情况;FCM法检测卡铂对细胞凋亡和细胞周期分布的影响;全波长荧光酶标仪检测细胞内活性氧自由基(ROS)水平变化。结果低剂量卡铂能诱导细胞衰老;不同浓度卡铂作用细胞48 h后,β-半乳糖苷酶染色阳性细胞显著增多;细胞周期多停滞于G0/G1期,S期显著减少,且细胞内ROS水平明显升高(P<0.05)。结论低剂量卡铂能诱导人肺癌细胞A549过早性衰老。
Objective To investigate the effect of low dose carboplatin on premature aging of lung cancer A549 cells. Methods A549 cells were treated with different concentrations of carboplatin. The inhibitory effect of carboplatin on cell proliferation was detected by MTT assay and clone formation rate assay. The cell senescence induced by carboplatin was detected by β-galactosidase staining assay. Cell apoptosis and cell cycle distribution. The changes of reactive oxygen species (ROS) levels in cells were detected by full wavelength fluorescence microplate reader. Results Low-dose carboplatin induced cell senescence. After treated with different concentrations of carboplatin for 48 h, the number of β-galactosidase positive cells increased significantly. The cell cycle was arrested in G0 / G1 phase and significantly decreased in S phase. ROS levels were significantly increased (P <0.05). Conclusion Low dose carboplatin can induce premature senescence of A549 human lung cancer cells.
