目的:评价“药食同源”中草药斑叶兰的各不同萃取部位对体外自由基的清除即抗氧化作用,并富集抗氧化活性组分从而制备乳膏制剂。方法:采用体外清除DPPH自由基的方法确定抗氧化活性最强的部位,通过HPLC外标法测定阿魏酸和香草酸是乙酸乙酯部位的主要成分,二者总含量占该部位的76.17%。以硅胶、凝胶柱层析法富集活性部位,并优化提取工艺和基质处方。结果:斑叶兰的乙酸乙酯部位清除自由基能力最为突出,在0.5 g·L~(-1)时达到92.31%,仅次于阿魏酸,IC_(50)分别为0.1622、0.1490 g·L~(-1)。结论:为将其研究开发成为天然中药美容护肤品奠定了基础,也为其开发为抗氧化药物用于治疗各种疾病提供了理论依据。
OBJECTIVE: To evaluate the anti-oxidative effect of various extraction sites of Chinese medicinal leaf spot leaves on the in vitro free radical scavenging activity and to enrich the antioxidant active components to prepare the cream preparation. METHODS: DPPH radical scavenging assay was used to determine the most active sites of antioxidant activity. Ferulic acid and vanillic acid were the main components of ethyl acetate by HPLC external standard method, accounting for 76.17% of the total. Silica gel, gel column chromatography enrichment of active site, and optimize the extraction process and matrix prescription. Results: The ability to scavenge free radicals in the ethyl acetate fraction of Pleurotus citri was the most prominent, reaching 92.31% at 0.5 g · L -1, second only to ferulic acid, with IC 50 of 0.1622 and 0.1490 g · L ~ (-1). Conclusion: It laid a foundation for its research and development into a natural herbal beauty skin care products, and provided a theoretical basis for the development of anti-oxidant drugs for the treatment of various diseases.
