目的 :观察过氧化氢诱导鼠白内障形成过程中晶状体上皮细胞NF κB的活化表达及葛根素 (Puerarin ,Pue)对NF κB活化表达的抑制作用 ,探讨NF κB在白内障发病过程中的意义及Pue对白内障的治疗作用。方法 :大鼠晶状体离体培养 ,分成过氧化氢组 (H2 O2 组 )、对照组、Pue处理组 (治疗组 ) ,免疫组织化学方法检测三组不同时间的晶状体上皮细胞NF κB的活化表达。结果 :随过氧化氢损伤时间的延长 ,H2 O2 组晶状体上皮细胞NF κB活化表达逐渐增强 ,与晶状体混浊程度正相关。培养 2 4小时 1mmol/l、10mmol/l浓度的Pue处理组与H2 O2 组比较晶状体上皮细胞NF κB平均阳性活化表达率和晶状体混浊程度均明显降低。结论 :过氧化氢可以诱导鼠晶状体上皮细胞NF κB的活化表达。 1mmol/l、10mmol/l浓度的Pue可以有效抑制NF κB的活化表达 ,有可能对抗或延缓白内障的发生。
OBJECTIVE: To observe the activation expression of NF-κB in lens epithelial cells induced by hydrogen peroxide and the inhibitory effect of Puerarin (Puerarin) on the activation of NF κB, and to explore the significance of NF κB in the pathogenesis of cataract and Pue Therapeutic effect of cataract. Methods: Rat lens were cultured in vitro and divided into H2O2 group, control group and Pue group (treatment group). The activation of NF κB in lens epithelial cells was detected by immunohistochemistry. Results: With H2O2 injury time prolonged, H2O2 group lens epithelial cells NF κB activation of expression gradually increased, and the degree of lens opacity was positively correlated. Compared with H 2 O 2 group, the average activation rate of NF κB and the degree of lens opacity in Puerarin group and H 2 O 2 group cultured for 24 h at 1 mmol / l and 10 mmol / l were significantly decreased. Conclusion: Hydrogen peroxide can induce the activation of NF κB in rat lens epithelial cells. Puerarin at a concentration of 1 mmol / l and 10 mmol / l can effectively inhibit the activation of NF κB, which may antagonize or retard the occurrence of cataract.
