目的:研究解毒活血通络中药复方防治动脉粥样硬化的作用机制及其效应靶点。方法:选取新西兰大耳白兔18只,随机分为3组,分别以生理盐水和解毒活血通络中药复方、阿托伐他汀连续灌胃7d,心脏采血,分离血清;体外培养人脐静脉内皮细胞,用脂多糖(LPS)刺激后,用含药血清干预,收集细胞,用Real-time PCR方法和Western Blot方法分别检测凝集素样氧化低密度脂蛋白受体-1(LOX-1)mRNA和蛋白表达、肿瘤坏死因子α(TNF-α)及细胞间黏附分子-1(ICAM-1)mRNA表达。结果:与空白对照组比较,用LPS刺激人脐静脉内皮细胞后,引起LOX-1 mRNA和蛋白、TNF-α、ICAM-1 mRNA的高表达(P<0.01),与模型组比较,用中药含药血清干预,显著抑制LOX-1mRNA和蛋白、TNF-α、ICAM-1 mRNA的高表达(P<0.01,P<0.05)。结论:解毒活血通络中药复方对LPS刺激后的人脐静脉内皮细胞具有保护作用,其机制与解毒活血通络中药复方对LOX-1、TNF-α和ICAM-1表达有抑制作用,减少单核细胞与血管内皮细胞黏附有关。
Objective: To study the mechanism and target of anti-atherosclerosis of traditional Chinese medicine of Jiedu Huoxuetongluo Recipe. Methods: Eighteen New Zealand white rabbits were randomly divided into three groups. The rats were randomly divided into three groups. Lectin-like oxidized low density lipoprotein receptor 1 (LOX-1) mRNA was detected by Real-time PCR and Western Blot And protein expression, tumor necrosis factor alpha (TNF-α) and intercellular adhesion molecule-1 (ICAM-1) mRNA expression. RESULTS: Compared with the blank control group, LOX-1 mRNA and protein, TNF-α and ICAM-1 mRNA were highly expressed in LPS-stimulated human umbilical vein endothelial cells (P <0.01) Serum-containing serum significantly inhibited LOX-1 mRNA and protein, TNF-α and ICAM-1 mRNA expression (P <0.01, P <0.05). Conclusion: Jiedu Huoxuetongluo Recipe has a protective effect on LPS-stimulated human umbilical vein endothelial cells. Its mechanism and detoxification Huoxuetongluo compound prescription can inhibit the expression of LOX-1, TNF-α and ICAM-1, Nuclear cells and vascular endothelial cell adhesion.
