目的 :研制具有乙酰胆碱酯酶 (AchE)活性的抗独特型抗体。方法 :AchEmAb是一种IgG1抗体 ,先用木瓜蛋白酶酶切 ,然后采用AchE Sepharose 4B和SPA亲和层析柱进行提取纯化 ,获得纯化的mAbFab段。以Fab作为抗原免疫小鼠 ,制备抗Fab片段的独特型抗体 (AIdAb)。结果 :酶催化ELISA法检测证明 ,抗mAb 3F3片段的AIdAb具有AchE活性。结论 :成功地制备了一株具有AchE活性的AIdAb ,为农药中毒的治疗开辟了新的途经
Objective: To develop an anti-idiotypic antibody with acetylcholinesterase (AchE) activity. Methods: AchE mAb, an IgG1 antibody, was digested with papain and then purified by AchE Sepharose 4B and SPA affinity chromatography to obtain a purified mAb Fab fragment. Mice were immunized with Fab as an antigen to prepare an anti-Fab fragment idiotypic antibody (AIdAb). Results: Enzyme-catalyzed ELISA demonstrated that AIdAb of anti-mAb 3F3 fragment had AchE activity. Conclusion: A strain of AIdAb with AchE activity was successfully prepared, which opened up a new way for the treatment of pesticide poisoning
