目的了解泰素和放射对人盲肠未分化腺癌细胞系(Hce8693)的联合作用效应。材料与方法应用MTT比色法,细胞核分裂指数,DNA琼脂糖电泳及流式细胞仪检测技术。结果泰素对该细胞的抑制呈一定的剂量依赖性;低浓度的泰素(10μg/ml)可致分裂细胞明显增加;单纯泰素(20μg/ml)和单纯放射作用后的18小时均产生细胞凋亡现象,而细胞经泰素联合放射作用后的3~24小时均出现了DNA梯状(Ladder)条带及凋亡(Apoptosis)小峰。结论泰素结合放射有一定协同作用,至少有相加效应。
Objective To understand the combined effects of paclitaxel and radiation on human cecal undifferentiated adenocarcinoma cell line (Hce8693). Materials and methods MTT colorimetry, nuclear index, DNA agarose electrophoresis and flow cytometry were used. Results Taxol had a dose-dependent inhibitory effect on the cells; low concentrations of Taxol (10 μg/ml) significantly increased the number of dividing cells, and simple Taxol (20 μg/ml) and 18 hours after irradiation alone produced Apoptosis was observed in cells, but ladders and apoptosis peaks appeared in 3 to 24 hours after cells were treated with taxol and combined radiation. Conclusions TAX combined with radiation has a certain synergistic effect, at least with additive effects.
