目的:研究在上海地区健康汉族人中不同年龄和性别的载脂蛋白E基因频率的群体分布.方法:本次调查包括649名健康汉族人(女:237人,男:412人).载脂蛋白E基因用PCR扩增,前引物:5′GGCACGGCTGTCCAAGGAGCT3′;后引物:5′GATGGCGCTGAGGCCGCGCT3′.PCR产物用5单位限制性内切酶CfoⅠ酶切,用20%聚丙烯酰胺凝胶电泳分离.结果:ApoE3频率最高为864%,其次ApoE4频率为75%,ApoE2频率最低为62%.经χ2检验,等位基因及基因型频率分布符合HardyWeinberg平衡.结论:本项研究提供了正常中国汉族人群中载脂蛋白E三个等位基因2,3和4频率的数据
OBJECTIVE: To study the population distribution of apolipoprotein E gene frequencies at different ages and genders among healthy Han Chinese in Shanghai. Methods: The survey included 649 healthy Han Chinese (237 females and 412 males). Apolipoprotein E gene amplified by PCR, the first primer: 5’GGCACGGCTGTCCAAGGAGCT 3 ’; after primer: 5’GATGGCGCTGAGGCCGCGCT 3’. The PCR product was digested with 5 units of restriction enzyme Cfo I and separated by 20% polyacrylamide gel electrophoresis. Results: The highest ApoE3 frequency was 86.4%, followed by ApoE4 at 75% and ApoE2 at 62%. By χ2 test, allele and genotype frequency distribution in line with the Hardy Weinberg equilibrium. CONCLUSIONS: This study provides data on the frequencies of the three alleles 2, 3, and 4 of apolipoprotein E in normal Chinese Han population
