目的分析长期饲喂n-3多不饱和脂肪酸(PUFA)高脂饮食对大鼠胰岛素敏感性的影响。方法雄性SD大鼠随机分为2组,对照组(NC)给予普通日粮,高脂组(TUF)给予富含n-3PUFA的高脂日粮。喂养16周,记录采食量及体质量,用高胰岛素—正常血糖钳夹技术评价大鼠胰岛素敏感性。结果 TUF组体质量与NC组没有统计学差异(P>0.05),基础血糖及胰岛素与对照组没有统计学差异(P>0.05),葡萄糖输注率(GIR)显著低于NC组(P=0.031)。结论长期过量摄入n-3PUFA导致大鼠胰岛素抵抗。
Objective To analyze the effect of long-term high-fat diet supplemented with n-3 polyunsaturated fatty acids (PUFA) on insulin sensitivity in rats. Methods Male Sprague-Dawley rats were randomly divided into two groups. The control group (NC) received normal diet and the high-fat diet group (TUF) received high-fat diet enriched with n-3PUFA. Fed for 16 weeks, the intake and body weight were recorded, and the insulin sensitivity of rats was evaluated by high insulin - normal blood glucose clamp technique. Results The body mass of TUF group was not significantly different from that of NC group (P> 0.05). There was no significant difference between baseline glucose and insulin in control group (P> 0.05) and glucose infusion rate (GIR) 0.031). Conclusion Long-term overexpression of n-3PUFA leads to insulin resistance in rats.
