目的研究木犀草素对人表皮共培养体系(黑素细胞与角质形成细胞)酪氨酸酶活性及黑素生成的影响。方法培养人表皮黑素细胞和人表皮黑素细胞与角质形成细胞共培养体系,利用四甲基偶氮唑蓝(MTT)还原法测定细胞活力,490nm比色法测定酪氨酸酶活性,475nm比色法测定黑素含量。结果 0.5μmol/L、0.75μmol/L和1μmol/L木犀草素对于黑素细胞及共培养细胞酪氨酸酶活性及黑素生成均有较强的剂量相关性增强作用,0.75μmol/L及1μmol/L木犀草素与对照组相比差异有统计学意义(P<0.01)。结论木犀草素对于共培养体系酪氨酸酶活性及黑素生成有较强的剂量相关性增强作用。
Objective To investigate the effects of luteolin on tyrosinase activity and melanogenesis of human epidermal coculture system (melanocytes and keratinocytes). Methods Human cortical melanocytes and human epidermal melanocytes were cultured with keratinocytes. MTT assay was used to determine the activity of tyrosinase, the activity of tyrosinase was determined by 490nm colorimetric assay, and the activity of tyrosinase at 475nm Colorimetric determination of melanin content. Results Luteolin treatment with 0.5μmol / L, 0.75μmol / L and 1μmol / L had a strong dose-dependent enhancement of tyrosinase activity and melanogenesis in melanocytes and co-cultured cells, and 0.75μmol / L and 1μmol / L luteolin compared with the control group, the difference was statistically significant (P <0.01). Conclusion Luteolin has a strong dose-dependent enhancement of tyrosinase activity and melanogenesis in co-culture system.
