利用脑内微透析技术与现代分析方法,考察了不同剂量川芎嗪对大鼠脑内多巴胺释放的影响。采用脑内微透析技术进行在体动态取样,建立了高效液相?电化学检测方法测定脑透析液中的多巴胺含量。结果显示川芎嗪皮下给药能够剂量相关性地增加大鼠脑内不同脑区中多巴胺的释放。该方法能够在不影响动物正常生理活动的情况下准确反映药物对大鼠脑内多巴胺释放量的影响,与传统神经递质研究方法相比,具有明显的优势。
Using the intracerebral microdialysis technique and modern analytical methods, we investigated the effects of different doses of ligustrazine on the dopamine release in rat brain. The intradial microdialysis technique was used to perform in-vivo dynamic sampling and a high performance liquid phase electrochemical detection method was established for the determination of dopamine in brain dialysate. The results showed that ligustrazine administered subcutaneously increased dopamine release in different brain regions in rat brain in a dose-dependent manner. The method can accurately reflect the effect of the drug on the dopamine release in the rat brain without affecting the normal physiological activity of the animal, and has obvious advantages compared with the traditional neurotransmitter research methods.
