目的探讨O139群、O1群霍乱弧菌分子遗传特征及其相互关系。方法用聚合酶链反应(PCR)、DNA序列分析及随机扩增多态性DNA(RAPD),检测了3株O139群、3株O1群ElTor生物型和2株古典生物型霍乱弧菌。结果3株O139群和5株O1群霍乱弧菌均扩增出566bp的DNA片段,DNA序列源于霍乱肠毒素(ctx)A2B基因。RAPD产生的两种DNA指纹图谱一致显示O139群与O1群ElTor生物型遗传特征极其相似,与古典生物型略有区别。结论O139群与O1群ElTor型存在密切的种系进化关系,前者可能由O1群ElTor生物型进化而来。
Objective To investigate the molecular genetic characteristics of Vibrio cholerae O139 and O1 strains and their relationship. Methods Three strains of O139, three strains of O1 ElTor and two strains of classical V. cholerae were detected by polymerase chain reaction (PCR), DNA sequence analysis and random amplified polymorphic DNA (RAPD). Results A total of 566bp DNA fragments were amplified from 3 O139 and 5 O1 Vibrio cholerae strains. The DNA sequence was from Cholera toxin (ctx) A2B gene. The two DNA fingerprinting profiles generated by RAPD consistently showed that the genetic characteristics of O139 group and O1 group ElTor were very similar, with a slight difference from the classical biological type. Conclusion There is close phylogenetic relationship between O139 and O1 ElTor. The former may be evolved from O1 ElTor.
