目的:建立一种可以快速提取和测定血清中的视黄醇的检测方法。方法:采用视黄醇乙酸酯作为内标,AgilentEclipse XDB-C18柱(5μm 4.6 mm×250 mm)液相色谱柱分离,甲醇-水(体积比98∶2)作为流动相,二级管阵列为检测器,反相色谱法测定血清中的视黄醇的含量进行检测。结果:样品加标回收率为88.2%~106.5%;视黄醇最低检出限为0.020μg/ml。结论:该方法具有简便、快速、准确的优点可用于血清中的视黄醇的检测。
OBJECTIVE: To establish a method for rapid extraction and determination of retinol in serum. Methods: The column was separated on a column of Agilent Eclipse XDB-C18 (5μm 4.6 mm × 250 mm) by using retinol acetate as the internal standard. The mobile phase was methanol-water (98: 2 by volume) As a detector, reverse phase chromatography was used to determine the level of retinol in serum. Results: The recoveries of spiked samples ranged from 88.2% to 106.5%. The detection limit of retinol was 0.020 μg / ml. Conclusion: This method is simple, rapid and accurate and can be used for the detection of retinol in serum.
