用PBS抽提、硫酸铵分段沉淀及猪甲状腺球蛋白—Sepharose 4B亲和层析可从腰点沙克沙(phaseolus vulgaris L)中分离得沙克沙凝集素,达到电泳纯。SDS—聚丙烯酰胺凝胶电泳得出一条带,分子量为30.26×10~3u,表明该凝集素由单一亚基组成。其含糖量为5%。该凝集素能凝集兔、小鼠、鸡、人A、B、O红细胞,其凝集作用可被猪甲状腺球蛋白明显抑制。该凝集素可有效地促进淋巴细胞转化和分裂;体外实验表明,它可凝集Sp 2/0骨髓瘤细胞。
Extraction with PBS, ammonium sulfate fractionation and porcine thyroglobulin-Sepharose 4B affinity chromatography can be used to isolate leflunomide from phaseolus vulgaris L to electrophoretic purity. SDS-polyacrylamide gel electrophoresis gave a band with a molecular weight of 30.26 × 10 ~ 3u, indicating that the lectin consists of a single subunit. The sugar content of 5%. The agglutinin can agglutinate rabbit, mouse, chicken, human A, B, O red blood cells, the agglutination can be significantly inhibited by porcine thyroglobulin. This lectin can effectively promote lymphocyte transformation and division; in vitro experiments show that it can agglutinate Sp 2/0 myeloma cells.
