[目的]观察绞股蓝提取液对自然衰老小鼠皮肤组织中过氧化氢酶(CAT)活性及丙二醛(MDA)含量的影响,并研究不同的给药途径对自然衰老小鼠皮肤组织的改善作用。[方法]昆明种3月龄雌性小鼠10只,作为青年对照组(A);昆明种12月龄雌性小鼠40只,随机分为4组,每组10只:老年外用空白组(B)、老年外用阳性对照组(C)、老年外用绞股蓝组(D)、老年(外用+内服)绞股蓝组(E)。A、B组外涂赋型剂0.5g/t,C组外涂维生素E20mg/t,D组外涂绞股蓝霜剂0.5g/t,E组外涂绞股蓝霜剂(剂量同D组),同时灌胃绞股蓝提取液0.8mL/t。30天后以生化分析方法测定皮肤组织中CAT活性、MDA含量,并观察皮肤组织结构改变。[结果]与A组比较,B、C、D、E组小鼠皮肤组织CAT活性均明显降低(P<0.01),MDA含量明显增加(P﹤0.01);与B组比较,C、D、E组CAT活性提高(P﹤0.05),MDA含量降低(P﹤0.01);与D组相比,E组CAT活性明显提高(P﹤0.05),MDA含量明显降低(P﹤0.05)。[结论]绞股蓝提取液具有拮抗小鼠皮肤自然衰老的作用;外用配以灌胃绞股蓝提取液该种给药方式能更好的起到延缓自然衰老对小鼠皮肤的损害。
[Objective] To investigate the effects of Gynostemma pentaphyllum extract on catalase (CAT) activity and malondialdehyde (MDA) content in skin of aging mice and to study the effects of different routes of administration on the skin tissue of mice with natural aging effect. [Method] Ten Kunming 3-month-old female mice were used as youth control group (A). Kunming 12-month-old female mice were randomly divided into 4 groups (n = 10) ), The positive control group (C), the external use of gypenosides (D), the elderly (external use + internal use) gypenotin group (E). 0.5g / t in group A and group B, 20mg / t in group C, 0.5g / t in group D, gypenosides cream in group E (same dose as group D) Gynostemma gavage 0.8mL / t. After 30 days, the CAT activity and MDA content in the skin tissue were determined by biochemical analysis method, and the skin tissue structure was observed. [Results] Compared with group A, the CAT activity in skin of mice in groups B, C, D and E were significantly decreased (P <0.01) and MDA content was significantly increased (P <0.01) Compared with group D, CAT activity in group E increased significantly (P <0.05) and MDA content decreased significantly (P <0.05). [Conclusion] Gynostemma extract could antagonize the natural aging of mice skin. The external application of Gynostemma pentaphyllum extract could effectively delay the skin damage caused by natural aging.
