目的 :初步探讨血管紧张素Ⅱ (angiotensinⅡ ,AngⅡ ) 1亚型受体 (AT1)拮抗剂洛沙坦 (Losar tan)对髂动脉球囊损伤后新生内膜的影响及其作用机制。方法 :家兔 36只 ,随机分为 3组 :对照组 (con组 ,n =1 3) ,大剂量洛沙坦组 (losH组 ,30mg/kg·d ,n =1 3) ,小剂量洛沙坦组 (losL组 ,7. 5mg/kg·d,n =1 0 )。 1周后髂动脉球囊损伤手术 ,测定血浆丙二醛浓度 (MDA) ,平均动脉压 (MAP)及术后 4周的血管内膜面积等指标 ,并进行少量电镜观察。结果 :losH组新生内膜面积较con组和losL组明显减小 (P <0 . 0 1 ) ;术后MDA较con组和losL组低 (P <0 . 0 5) ;losH组和losL组MAP均较con组明显降低 (P <0 . 0 5) ;losH组新生内膜的血管平滑肌细胞 (VSMC)以收缩型为主 ,con组以分泌型为主。结论 :洛沙坦对兔髂动脉球囊损伤所致的内膜增生有抑制作用 ,其机制似与降低血压无关
Objective: To investigate the effect and mechanism of Losar tan, an angiotensin Ⅱ (AngⅡ) 1 subtype receptor (AT1) antagonist, on neointima formation after balloon injury in iliac arteries. Methods: Thirty-six rabbits were randomly divided into three groups: control group (con group, n = 13), high dose losartan group (losH group, 30mg / kg · d, n = 13) Saxan group (losL group, 7.5 mg / kg · d, n = 10). One week later, balloon injury of iliac arteries was performed to measure plasma MDA, mean arterial pressure (MAP) and intima area at 4 weeks postoperatively. Results: The neointimal area in losH group was significantly lower than that in con group and losL group (P <0.01). The MDA level in losH group was lower than that in con group and losL group (P <0.05) MAP was significantly lower than con group (P <0. 05); losH group of neointimal smooth muscle cells (VSMC) to contraction type, con group mainly secretory type. Conclusion: Losartan can inhibit intima hyperplasia induced by rabbit balloon iliac artery injury, and its mechanism seems to be independent of hypotension
