目的:研究岩乌头根部的二萜生物碱类成分及其抗PAF活性。方法:95%乙醇提取,所得浸膏采用经典的酸-碱处理方法,得到总碱,总碱经硅胶,RP-18,Sephadex LH-20等多种材料柱色谱分离,得到生物碱单体,再通过波谱解析鉴定其化学结构;并对分离鉴定的生物碱进行抗PAF活性的检测。结果:分离鉴定了5个二萜类生物碱,分别为sachaconitine(1),14-acetylsachaconitine(2),hemsleyanineC(3),circinasineA(4),talatisamine(5);化合物1和2显示了一定的抗PAF活性。结论:所有化合物均为首次从该植物中分得,化合物1和2对PAF诱导的血小板聚集具有一定的抑制作用。
Objective: To study the components of diterpene alkaloids and their anti-PAF activity in the root of Aconitum kusnezoffii. Methods: The extract was extracted with 95% ethanol. The obtained extract was treated with classical acid-alkali method to get the total alkaloids. The total alkaloids were separated by column chromatography on silica gel, RP-18, Sephadex LH- And then identified by chemical analysis of its chemical structure; and alkaloids isolated and tested for anti-PAF activity. Results: Five diterpenoid alkaloids were isolated and identified as sachaconitine (1), 14-acetylsachaconitine (2), hemsleyanine C (3), circinasine A (4) and talatisamine Anti-PAF activity. CONCLUSION: All compounds were isolated from this plant for the first time. Compounds 1 and 2 inhibited PAF-induced platelet aggregation to some extent.
