应用细胞周期流式细胞仪(FCM)分析与~3H—TdR掺入法药敏测定技术,对8种化疗药物(MMC,MTX,5—Fu,ADM,CDDP,VP_(16),VCR,DTIC)在29例肝癌体外培养细胞的抑制作用与细胞周期比例的关系进行了对比研究。结果显示,化疗药物抑制率与细胞周期比例有关。以抑制率>60%作为有效标准,则对8种化疗药物有效的22例肝癌(75.9%)的细胞周期比例明显不同于抑制率<60%的肝癌。我们认为,FCM细胞周期测定具有简便、快速、客观、准确的特点,在临床个体化疗上具有实际应用价值。
Cytokine Flow Cytometry (FCM) Analysis and ~3H-TdR Incorporation Method for Drug Sensitive Determination of 8 Chemotherapeutic Drugs (MMC, MTX, 5-Fu, ADM, CDDP, VP_(16), VCR, DTIC ) A comparative study was performed on the relationship between the inhibitory effect of 29 hepatocellular carcinoma cells in vitro and the cell cycle ratio. The results showed that the inhibitory rate of chemotherapeutic drugs was related to the cell cycle ratio. With an inhibition rate of >60% as an effective criterion, the cell cycle proportion of 22 cases of hepatoma (75.9%) effective for 8 chemotherapeutic agents was significantly different from that of liver cancer with inhibition rate <60%. We believe that the FCM cell cycle assay is simple, rapid, objective and accurate, and has practical application value in clinical individual chemotherapy.
