补肾活血中药对大鼠慢性高眼压模型初级视皮质BDNF损害的影响(英文)

目的:观察补肾活血中药对大鼠慢性高眼压(elevated intraocular pressure,EIOP)模型初级视皮质(primary visual cortex,PVC)脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)的干预作用,并对其作用机理进行初步探讨。方法:采用烙闭上巩膜静脉法,烙闭大鼠3支上巩膜静脉,制作大鼠慢性EIOP模型,随机分为3组:模型组,给药组,空白组。连续灌胃8wk,并于8wk末处死大鼠,观察其对EIOP大鼠眼压、PVC的BDNF表达及神经元细胞超微结构的影响。结果:本实验采用的烙闭上巩膜静脉的造模方法使大鼠眼压明显升高(P<0.01),与造模前比较差异有显著统计学意义(P<0.01);PVC病理切片半定量分析表明,模型组BDNF总面积82438±2597.39(S/μm2)、平均光密度1155.9±123.14、积分光密度12915±673.28,与空白组(总面积132370±7588.47S/μm2,平均光密度5365±379.65,积分光密度35102±2648.5)比较均有显著统计学意义(均为P<0.05),模型组BDNF总面积与给药组(108980±9126.77S/μm2)比较有统计学意义(P<0.05),给药组平均光密度(3220.4±413.67)与模型组比较有统计学意义(P<0.05),给药组积分光密度(23821±3431.68)与模型组比较,差异有统计学意义(P<0.05)。结论:补肾活血中药通过增强BDNF的表达、改善神经元细胞超微结构而促进EIOP大鼠初级视皮质损伤的修复。 Objective: To observe the effects of Bushen Huoxue Chinese medicine on the intervention of primary visual cortex (PVC) brain-derived neurotrophic factor (BDNF) in rats with elevated intraocular pressure (EIOP) And its mechanism of action to conduct a preliminary discussion. Methods: The model of chronic EIOP was made by means of the method of branching and closing the sclera vein and the scleral vein of 3 rats in the rats. The rats were randomly divided into 3 groups: model group, administration group and blank group. The rats were sacrificed at the end of the 8th week, and their effects on the intraocular pressure, expression of BDNF in PVC and the ultrastructure of neurons in EIOP rats were observed. Results: The intraocular pressure (IOP) was significantly increased in this experiment (P <0.01), which was significantly different from that before model establishment (P <0.01). The pathological section of PVC Quantitative analysis showed that the total area of ​​BDNF in the model group was 82438 ± 2597.39 (S / μm2), the average optical density was 1155.9 ± 123.14, and the integral optical density was 12915 ± 673.28. Compared with the blank group (total area 132370 ± 7588.47S / μm2, average optical density 5365 ± (P <0.05). The total area of ​​BDNF in the model group was significantly higher than that in the administration group (108980 ± 9126.77S / μm2) (P <0.05), and the integral optical density was 35102 ± 2648.5 ) (P <0.05). The mean optical density of the treated group (3220.4 ± 413.67) was significantly higher than that of the model group (P <0.05). The integral optical density of the treated group (23821 ± 3431.68) was significantly lower than that of the model group <0.05). Conclusion: Bushen Huoxue medicine can promote the repair of primary visual cortex injury in EIOP rats by enhancing the expression of BDNF and improving the ultrastructure of neurons.
