本文报导用酸性α-醋酸萘酚酯酶(ANAE)染色法检查新生期去胸腺小鼠外周血T细胞的变化.正常C_(57)BL小鼠外周血ANAE~+淋巴细胞比例在生后0、3、6、14和成年组分别为5.6%、19.49%、26.7%和50.2%.在生后0、3、6天去胸腺组和模拟手术组小鼠2周龄时则分别为4.5%、7.6%、17.1%和35.0~41.0%.另外,胸腺残留组小鼠的ANAE~+淋巴细胞百分率明显高于去胸腺组.这些结果证实小鼠外周血中ANAE~+淋巴细胞主要是来自胸腺的T细胞.新生期去胸腺小鼠注射猪或牛胸腺素F5(每天200ug/只)12天后,外周血ANAE~+淋巴细胞百分率由原来的9.8~14.5%提高到21.3~28.2%.对照组无明显增加,提示T细胞的ANAE活性与胸腺素有关.作者提出ANAE染色法是鉴定新生期去胸腺小鼠模型的可靠指标,也可用于测定胸腺素的活性.
This article reports the change of peripheral blood T cells in neonatal rats with thymic deprivation using acid alpha-naphthol esterase (ANAE) staining method. The ratio of ANAE~+ lymphocytes in peripheral blood of normal C_(57)BL mice is 0 , 3, 6, 14 and adult groups were 5.6%, 19.49%, 26.7%, and 50.2%, respectively. In the thymus group at 0, 3, and 6 days after birth and the mice in the simulated surgery group were 4.5% at 2 weeks of age. , 7.6%, 17.1%, and 35.0-41.0%. In addition, the percentage of ANAE~+ lymphocytes in the thymus-remnant group was significantly higher than that in the dethymus group. These results confirmed that the ANAE~+ lymphocytes in the peripheral blood of mice were mainly derived from the thymus. The number of ANAE~+ lymphocytes in the peripheral blood increased from 9.8 to 14.5% to 21.3 to 28.2% after 12 days of thymic mice injected with porcine or bovine thymosin F5 (200ug/day). No significant increase, suggesting that the T cell ANAE activity is related to thymosin. The authors proposed that the ANAE staining method is a reliable indicator to identify neonatal dethymic mouse models, can also be used to determine the activity of thymosin.
