利用NC裸鼠复制人鼻咽癌(NPC)动物模型和低分化鼻咽癌细胞株(CNE_2)培养,检测不同时期荷人NPC裸鼠血浆MDA,以探讨荷瘤鼠血中自由基的变化规律和肿瘤的关系,并以CNE_(-2)细胞株为线索,检测CNE_(-2)细胞株培养上清液MDA,间接追踪荷人NPC裸鼠血中自由基变化的来源。结果表明荷人NPC裸鼠潜伏期血浆MDA已明显高于正常裸鼠(P<0.01),且荷人NPC裸鼠血浆中MDA从早期到中期逐渐升高,并达高峰,中期至晚期MDA渐下降,但仍高于正常水平(P<0.01); CNE_(-2)细胞株培养上清液中MDA明显高于人胚肺细胞株培养上清液所含MDA(P<0.01),并随着培养时间延长而增多。提示肿瘤发展与自由基水平的增高有关,荷人NPC裸鼠血浆MDA增高可能来源于肿瘤细胞自由基产生过多或清除不足。
Human nasopharyngeal carcinoma (NPC) animal model and nasopharyngeal carcinoma cell line (CNE_2) cultured in NC nude mice were used to detect plasma MDA in NPC nude mice at different stages to explore the changes of free radicals in the blood of tumor-bearing mice. In relation to tumors, the CNE_(-2) cell line was used as a clue to detect the MDA in the culture supernatant of the CNE_(-2) cell line. The source of free radical changes in the blood of the NPC nude mice was directly traced. The results showed that the plasma MDA in nude mice bearing human NPC was significantly higher than that in normal nude mice (P<0.01), and the MDA in the nude mice bearing human NPC gradually increased from early to mid-term and reached a peak, and the MDA in the middle to late stage decreased. , but still higher than normal level (P<0.01); MDA in the culture supernatant of CNE_(2) cell line was significantly higher than that in human embryo lung cell line culture supernatant (P<0.01), and with The cultivation time has increased and increased. It is suggested that the development of tumors is related to the increase of free radicals. The increase of plasma MDA in NPC nude mice may be due to excessive generation of free radicals or insufficient clearance of tumor cells.
