目的 检测所构建的沙眼衣原体 (Ct)重组疫苗株口服免疫小鼠后 ,产生的体液免疫应答 .方法 将所构建的重组菌以 5× 10 8菌落形成单位 (CFU ) /只的剂量 ,口服免疫雌性 BAL B/ c小鼠 ,随机分为 7组 ,每组 3只 .于免疫后第 2 ,7,14,2 1,2 8,35和 42 d各取 1组 ,采集血清、小肠肠腔冲洗液和阴道冲洗液标本 .分别以 D型 Ct和鼠伤寒杆菌临床分离株为抗原 ,检测各组血清、小肠冲洗液和阴道冲洗液标本中特异性 Ig G,Ig A和 Ig M.取抗 D型 Ct血清 Ig G阳性标本和阴道冲洗液 Ig A阳性标本 ,分别用于检测与 C型 Ct的交叉反应 .结果 在血清标本中 ,有抗 D型 Ct和抗鼠伤寒杆菌特异性 Ig G和 Ig M,在小肠冲洗液和阴道冲洗液标本中有抗上述两种抗原特异性 Ig G和 Ig A.抗 D型 Ct Ig G阳性血清标本和 Ig A阳性阴道冲洗液标本与 C型 Ct均有交叉反应 .结论 所构建的疫苗株免疫小鼠后 ,能够诱导 Ct特异性 Ig G及 Ig A产生 .
Objective To detect the humoral immune response of the constructed Ct recombinant vaccine strain after oral immunization in mice.Methods The constructed recombinant strains were orally immunized with 5 × 10 8 colony forming units (CFU) per mouse Female BAL B / c mice were randomly divided into 7 groups with 3 mice in each group.At 1, 2, 7, 14, 2, 12, 8, 35 and 42 d after immunization, Rinse solution and vaginal rinse solution.Methods D-type Ct and Salmonella typhimurium clinical isolates were used as the antigen to detect the specific Ig G, Ig A and Ig M in serum, small intestinal rinse and vaginal rinse D type Ct serum Ig G positive specimens and vaginal wash Ig A positive specimens were used to detect cross-reaction with C-type C. Results In serum samples, anti-D-Ct and anti-Salmonella typhimurium-specific Ig G and Ig M against the two antigen-specific Ig G and Ig in both intestinal and vaginal wash samples A. Anti-D-type Ct Ig G positive serum samples and Ig A-positive vaginal wash samples and C-type Ct were Cross-reaction.Conclusion The constructed vaccine strains can induce Ct-specific immunity in mice Heterosexual Ig G and Ig A production.
