目的探讨胃癌RUNX3基因表达对细胞凋亡与增殖细胞转换[凋亡率与增殖率之比值(AI/PI)]的影响及其与胃癌预后的关系。方法应用免疫组织化学分析RUNX3基因的表达;应用流式细胞术分析胃癌细胞的凋亡率和增殖率;采用Kaplan-Meier限乘法计算生存率。结果在60例胃癌中RUNX3基因的阳性表达率为46.7%。胃癌细胞RUNX3基因表达与淋巴转移和远处转移有关(P<0.05)。胃癌细胞的AI/PI经频数分布表分析和检验均服从正态分布。胃癌细胞AI/PI范围显示为0.05~0.46,胃癌细胞AI/PI与淋巴转移和远处转移有关(P<0.05)。胃癌细胞中RUNX3和AI/PI的表达之间存在明显相关关系(r=0.39,P<0.05)。用Kaplan-Meier生存曲线经Logr-ank检验发现,RUNX3的阳性表达与生存相关(P<0.05)。结论RUNX3基因可能通过影响凋亡与增殖细胞转换而影响胃癌的发生发展;RUNX3基因检测可作为胃癌临床病理学特点和预后的指标。
Objective To investigate the effect of RUNX3 gene expression on the apoptosis and proliferative cell cycle (AI / PI ratio) in gastric cancer and its relationship with the prognosis of gastric cancer. Methods The expression of RUNX3 gene was analyzed by immunohistochemistry. The apoptosis rate and proliferation rate of gastric cancer cells were analyzed by flow cytometry. The survival rate was calculated by Kaplan-Meier method. Results The positive rate of RUNX3 gene was 46.7% in 60 cases of gastric cancer. The expression of RUNX3 in gastric cancer cells was correlated with lymphatic metastasis and distant metastasis (P <0.05). Gastric cancer AI / PI frequency distribution table analysis and testing are subject to normal distribution. The range of AI / PI of gastric cancer cells was 0.05-0.46, and AI / PI of gastric cancer cells was related to lymph node metastasis and distant metastasis (P <0.05). There was a significant correlation between RUNX3 and AI / PI expression in gastric cancer cells (r = 0.39, P <0.05). Log-rank test with Kaplan-Meier survival curve showed that the positive expression of RUNX3 correlated with survival (P <0.05). Conclusion RUNX3 gene may affect the occurrence and development of gastric cancer by influencing apoptosis and proliferation of cells. The detection of RUNX3 gene may be used as an indicator of clinicopathological features and prognosis of gastric cancer.
