目的探讨FK409对SD大鼠缺血再灌注后心肌的影响。方法将SD大鼠随机分为四组,空白对照组、FK409低剂量组、中剂量组和高剂量组,均进行缺血再灌注模型制备。空白对照组在造模后5min静滴生理盐水至实验结束,低剂量组、中剂量组和高剂量组分别在进行造模后5min给予FK409 0.1nmol/kg、0.5nmol/kg和1.0nmol/kg。用多道生理记录仪持续监测大鼠心电图机血流动力学指标(HR、LVDP、±dP/dt max),并抽取大鼠血液进行CK、LDH、SOD、NO、H2O2、MAD等指标的检测。数据均进行记录并采用SPSS13.0进行统计学分析。结果 FK409对SD大鼠缺血再灌注后心功能指标均有所改善,中剂量组改善最明显,与其他组比较差异有统计学意义(P<0.05)。实验指标CK、LDH、SOD、NO、H2O2、MAD也均有所改善,除NO较对照组升高外,其余指标均有一定程度的下降。其中,以中剂量组最明显与其他各组比较差异均具有统计学意义(P<0.05)。结论一定剂量FK409在SD大鼠心肌缺血再灌注中可以提高NO含量从而降低缺血再灌注后血清中CK、LDH、SOD、H2O2、MDA等指标,改善缺血再灌注后SD大鼠心肌细胞的损伤。
Objective To investigate the effect of FK409 on myocardium after ischemia-reperfusion in SD rats. Methods SD rats were randomly divided into four groups. The blank control group, FK409 low dose group, middle dose group and high dose group were all prepared for ischemia reperfusion. The rats in the blank control group were injected with normal saline 5 min after the model was established, and the rats in the low dose group, middle dose group and high dose group were given 0.1, 0.5 and 1.0 nmol / kg of FK409 . The indexes of hemodynamics (HR, LVDP, ± dP / dt max) in the electrocardiogram of rats were continuously monitored with a multi-channel physiology recorder, and the indexes of CK, LDH, SOD, NO, H2O2 and MAD . Data were recorded and analyzed by SPSS 13.0. Results FK409 improved cardiac function after ischemia-reperfusion in SD rats, with the most obvious improvement in middle-dose group. The difference was statistically significant (P <0.05). The indexes of CK, LDH, SOD, NO, H2O2 and MAD also improved. Except for the increase of NO compared with the control group, the other indexes all decreased to a certain extent. Among them, the difference between the most obvious dose group and the other groups was statistically significant (P <0.05). Conclusions FK409 can increase the content of NO and decrease the levels of CK, LDH, SOD, H2O2 and MDA in SD rats after myocardial ischemia / reperfusion, and improve myocardial cell apoptosis after myocardial ischemia / reperfusion Damage.
