利用环孢菌素A引起棒曲霉SIA-F4770形态的特异变化,建立筛选具有免疫抑制活性抗真菌剂的琼脂平板法。并用已知抗生素证实了方法的有效性。该方法不仅可根据检测菌不同的形态变异定向筛选具有免疫抑制活性和抗真菌活性的微生物代谢产物,而且具有简单、快速、选择性高和不受代谢产物中其他抗真菌组分干扰等优点。用该方法过筛4500个真菌代谢产物粗品,从中筛选到环孢菌素A的已知产生菌、环孢菌素C的新产生菌和多种青霉产生的共同代谢产物brefeldinA。
Cyclosporine A was used to induce the specific morphological changes of Aspergillus clavatus SIA-F4770, and an agar plate method for screening antifungal agents with immunosuppressive activity was established. The effectiveness of the method was confirmed with known antibiotics. The method not only can screen microorganism metabolites with immunosuppressive activity and antifungal activity according to different morphological variations of the tested bacteria, but also has the advantages of being simple, rapid, highly selective and not disturbed by other antifungal components in metabolites. Using this method, 4,500 crude fungal metabolites were screened, and the known bacterium of cyclosporin A, new bacterium of cyclosporin C and brefeldin A, a common metabolite produced by various Penicillium strains were screened out.
