无色花色素还原酶(leucoanthocyantin reducase,LAR)是植物类黄酮生物合成途径中的一个关键酶。本研究根据海南龙血树(Dracaena cambodiana)转录组数据,利用RT-PCR技术克隆1个编码无色花色素还原酶基因DcLAR1,结果表明:该基因全长1 355 bp,包含一个1 011bp的开放阅读框,编码336个氨基酸。DcLAR1蛋白具有典型的植物无色花色素还原酶结构特征,包含3个保守的结构域RFLP、ICCN和THD。表达分析结果表明,DcLAR1在根、茎、花、叶和果中具有不同程度的表达;DcLAR1表达受到血竭诱导剂的诱导,与血竭积累正相关,推测该基因可能在海南龙血树类黄酮生物合成过程中具有重要功能。
Leucoanthocyantin reductase (LAR) is a key enzyme in the biosynthesis pathway of plant flavonoids. In this study, we cloned a gene encoding DcLAR1, a colorless anthocyanidin reductase gene, from Dracaena cambodiana transcriptome data. The results showed that the gene was 1 355 bp in length and contained a 1 011 bp open Reading frame, encoding 336 amino acids. The DcLAR1 protein has typical structural features of plant leucovorin reductase, including three conserved domains RFLP, ICCN and THD. The results of expression analysis showed that DcLAR1 expressed differently in roots, stems, flowers, leaves and fruits. The expression of DcLAR1 was induced by DRG and positively correlated with the accumulation of DRx, suggesting that DcLAR1 might be expressed in the species of Dracaena Flavonoids have important functions in biosynthesis.
