应用PHA、ConA刺激经IFN-α,rIFN-γ作用24小时后的正常人外周血单个核细胞,观察其对产生IL-2、IF N-γ能力与环核苷酸含量的影响。结果表明rFN-α、IFN-γ对单个核细胞产生IL-2、IFN-γ能力的影响取决于外源性IFNs的剂量,随作用的IFN-α、rIFN-γ剂量增加,渐出现抑制性效应。IFNs作用后的单个核细胞诱生IL-2、IFN-γ能力之间有较好的相关关系,提示它们关系密切。此外,IFN-α,rIFN-γ能够增加单个核细胞内cAMP含量与cAMP/cGMP比值,提示这一作用可能同IL-2、IFN-γ等免疫调节因子的细胞生物学调节效应有关。
The effects of IFN-α and rIFN-γ on the production of IL-2, IF N-γ and the contents of cyclic nucleotides in normal human peripheral blood mononuclear cells after stimulated by IFN-α and rIFN-γ were detected by PHA and ConA. The results showed that the effect of rFN-α and IFN-γ on the production of IL-2 and IFN-γ by mononuclear cells was dependent on the dose of exogenous IFNs. With the increase of IFN-α and rIFN-γ doses, the inhibition appeared effect. There was a good correlation between IL-2 and IFN-γ induced by IFNs after mononuclear cells, suggesting that they are closely related. In addition, IFN-α and rIFN-γ can increase the ratio of cAMP to cAMP / cGMP in mononuclear cells, suggesting that this effect may be related to the regulation of cell biology of immunomodulatory factors such as IL-2 and IFN-γ.
